实验性自发性高血压大鼠脑出血急性期中枢及外周血管紧张素受体的表达与血压调控的相关性

Study of the Relationship Between Blood-Pressrue Regulation and Angiotensin Receptors in Center and Periphery after Acute Intracerebral Hemorrhage in Spontaneously Hypertensive Rats

摘要: 目的 通过观察自发性高血压大鼠(Spontaneously hypertensive rats,SHR)脑出血急性期血压变化和中枢及外周血管紧张素受体(angiotensin receptors,ATR)mRNA及蛋白的表达情况,探讨脑出血急性期血压调控与ATR的相关性。方法 选取6月龄雄性SHR大鼠,将胶原酶注入尾壳核,建立脑出血(Intracerebral Hemorrhage,ICH)模型,使用尾袖法监测血压变化;运用免疫组化法和反转录聚合酶链反应(Reverse-Transcription Polymerase Chain Reaction,RT-PCR)法检测出血后各时间点脑及肾脏组织与对照组ATR mRNA及蛋白表达情况。结果 (1)SHR大鼠脑出血第1天血压明显升高,与出血前相比较差异具有统计学意义(P<0.05),第3天明显下降,第7~14天基本恢复至出血前水平;(2)脑组织中AT1R表达上调,第3天达高峰,与出血前比较差异具有统计学意义(P<0.05),第7~14天基本恢复至出血前水平;(3)脑组织中的AT2R和肾脏中的AT1R、AT2R各时间点表达量无统计学差异(P>0.05)。结论 SHR脑出血急性期血压调控可能与选择性上调脑组织中的AT1R表达有一定的相关性。

Abstract: Objective To study the relationship between ATR and blood-pressrue regulation. Methods A total of 30 six-month-old male SHR were used, The ICH model was established by infusing collagenase into rat caudate nucleus. Monitoring the change of blood pressure by tail-cuff. Immunohistochemistry and Reverse-Transcription Polymerase Chain Reaction(RT-PCR) were performed to detect the expression of ATR in center and periphery after ICH. Results (1) the blood pressure in SHR significantly increased after ICH at the first day(P<0.05), decreased at the third day and recovered from the seventh day to fourteenth day; (2) expression of AT1R in the brain up-regulated after ICH, especially at the third day(P<0.05), and then recovered from the seventh day to fourteenth day, (3) AT2R in the brain as well as AT1R and AT2R in the kidney no difference were found after ICH. Conclusion Blood pressure increased after ICH in SHR might be related with selectivity up-regulation AT1R in the brain.
