线栓法制备大鼠局灶性脑缺血/再灌注模型的改良及探讨

Improvement and Exploration on Local Cerebral Ischemia/Reperfusion Model in Rats by Intraluminal Suture Technique

摘要:

【摘要】 目的 探讨建立一种操作简便、稳定性好、成功率高的大鼠局灶性脑缺血/再灌注模型。 方法 60只清洁级成年健康Wistar雄性大鼠,随机分为传统组和改良组各30只,传统组参照Zea Longa法制备大鼠局灶性脑缺血/再灌注模型,改良组主要针对三个方面进行改良以提高成功率,涉及线栓头端的处理、插入线栓的方式以及插入线栓位置的选择等。通过神经功能评分及脑组织学检查来验证模型稳定性和成功率。 结果 制模后改良组(2.35±1.23)与传统组(1.24±1.50)神经功能评分相比差异有显著性(t=2.852;P=0.007)。改良组实验动物模型制作成功率(63.33%)、存活动物模型制作成功率(73.08%),与传统组(20.00%和33.33%)相比较差异均有显著性(χ2值分别为11.589和6.848,P值分别为0.001和0.009)。 结论 改良组模型神经功能评分、存活动物模型制作成功率和实验动物模型制作成功率均高于传统组,是较为理想的制备大鼠局灶性脑缺血/再灌注模型的方法。

Abstract:

【Abstract】 Objective To establish a simple, reliable and stabilized rat model of local cerebral ischemia/reperfusion model in rats. Methods The local cerebral ischemia/reperfusion model in rats was set up by referring to Zea Longa method. Sixty Wistar rats were randomly divided into 2 groups(30 cases in modified group, 30 cases in traditional method group). Modified group mainly focused on three aspects to improve the success rate, involved the treatment of the line bolt head, method of how to insert the intraluminal suture and selection of insertion position etc. Stability and the success rate of model were detected by behavioral test of neurological deficit scoring, magnetic resonance imaging(MRI) inspection and pathological examination. Results Neurological function score of the modified group(2.35±1.23) points had significant difference compared with the traditional group(1.24±1.50) points(t=2.852, P=0.007), the success rate of survival animal model and experimental animal model were different significantly between the modified group(73.08%, 63.33%) and traditional group(33.33%, 20.00%)(χ2=11.589 & 6.848, P=0.001 & 0.02). Conclusions Neurological scores and success rate of modified group model was significantly higher than the traditional group. It is an ideal making model approach of local cerebral ischemia/reperfusion.
